Öğe Delayed diagnosis in a mentally motor-retarded girl: A rare case of giant mucinous cystadenoma(2023) Tuşat, Mustafa; Genç, Şerife ÖzlemThis presentation discusses the challenging case of a 15-year-old mentally motor-rarity of mucinous cystadenomas in childhood, with only ten reported cases in the literature. Case Report: We present a 15-year-old MMR girl who has been bedridden since birth. She visited the emergency department three times within two days with complaints of severe vomiting, restlessness, and abdominal swelling. Initially, she was misdiagnosed with constipation and sent home with incorrect treatment. An emergency abdominal CT scan revealed a 13x7x7 cm cystic lesion in the left ovary, filling the abdominal cavity (Figure 1a-e). Left salpingo-oophorectomy was performed, confirming a mucinous cystadenoma (Figure 2,3). Conclusion: This complex case underscores the importance of considering ovarian masses in children with abdominal symptoms and the significance of timely imaging. Hsp70 is a molecular chaperone with physiological roles such as folding of newly synthesized proteins, refolding of misfolded proteins, and inhibition of protein aggregation [2]. Although it is expressed at low levels in healthy cells, Hsp70 is overexpressed in tumor cells and helps tumor cells survive in hard conditions. High levels of Hsp70 play a role in many processes related to poor prognosis, such as inhibition of apoptosis in tumor cells, resistance to chemotherapy, metastasis, and invasion. Therefore, specific inhibition of Hsp70 in tumor cells is an important strategy in cancer therapy. In addition, while inhibition of Hsp70 has toxic effects on the tumor, its absence of toxic effects on normal cells is very important in preventing systemic toxicity in cancer therapy [3]. This study, it was aimed to investigate the cytotoxic effect of Hsp70 inhibitor JG-98 on K562 chronic myeloid leukemia cells. For this purpose, human chronic myeloid leukemia K562 cells were cultured and treated with different concentrations (100-0.1 µM) of JG-98 inhibitor. Cell viability was evaluated by XTT method after 24 and 48 hours of incubation. Cell viability concentration was found by accepting the cell viability of the control group as 100%, and accordingly, JG-98 was found to be effective even at low doses. The findings showed that the Hsp70 inhibitor JG-98 significantly inhibited K562 cell proliferation (p<0.05). The IC₅₀ values were calculated as 6.37 µM and 2.39 µM, respectively, after 24 and 48 hours of the incubation period. The study shows that JG-98 promises in the treatment of chronic myeloid leukemia.Öğe 6 IN VITRO ANTIPROLIFERATIVE ACTIVITIES OF ROYAL JELLY COLLECTED FROM SIVAS IN HUMAN OVARIAN CANCER (SK-OV-3) CELL LINE(2022/9/17) Güzel, Mehmet Hakan; Daştan, Taner; Durna Daştan, Sevgi; Yurtcu, NazanRoyal jelly has positive effects on human health. It is a bee product and functional food. Royal jelly is secreted from the glands of hypopharyngeal and mandibular secretion tissue of young worker bees. It is a viscous liquid with a sour taste and a white-yellowish color and has a special phenol smelling liquid. Royal jelly is health promoting food, and it is used in the production of cosmetic medical products. Most of the study in the literature, it has been shown that royal jelly has antibacterial, anti-inflammatory, vasodilator, hypotensive, antioxidant, antidiabetic, antihypercholesterolemic and antitumoral activities [1]. Within the aim of this study, royal jelly samples were obtained from various localities of Sivas province. Royal jelly samples were obtained from beekeepers in the 2020 harvest period. It was aimed to determine and compare the cytotoxic activity of royal jelly samples collected different area of Sivas province in SK-OV-3 (Human ovarian cancer) cell line using MTT test. MTT results were determined spectrophotometrically and IC50 values were calculated with Graphpad program. It was observed that some samples with a high concentration dosage and during 48 hour period showed good cytotoxic activity in this cell line. It is the first time to test the efficacy of royal jelly samples from Sivas region on the SKOV-3 cell line and this study will be a beginning in terms of the therapeutic use of bee royal jelly samples from Sivas province and the preparation of new effective supplements.Öğe Polikistik Over Sendromlu Hastalarda Obezite ve Lipid Profilinin Karotis İntima Kalınlığı ile İlişkisi(2022/11) Yurtcu, Nazan; KARADEMİR, Dilay; SERİN SercanAmaç Polikistik over sendromu (PKOS), Üreme çağındaki kadınlarda önemli bir reprodüktif bozukluk olup, metabolik rahatsızlıklar ve kardiyovasküler hastalıklara da yol açabilmektedir. Bu çalışma, PCOS hastalarında Karotis intima media kalınlığı (KİMK)'na antropometik ölçümler, kan hormon değerleri ve lipid profilinin etkisinin değerlendirilmesi amaçlandı. Yöntem Bu prospektif çalışmada, PKOS tanısı almış olan ve çalışmaya dahil edilme/dışlanma kriterlerini karşılayan 108 gönüllü ve 92 sağlıklı, aynı yaş aralığında olan kadınlar kontrol grubu olarak çalışmaya dahil edildi. Çalışmaya alınan PKOS ve kontrol grubu katılımcılar obez olan ve olmayan olarak kendi içerisinde iki alt grubuna ayrıldı. Çalışmaya alınan tüm kadınlardan yaş, vücut kitle indeksi, bel çevresi, hormonal ve lipid değerleri elde edildi. Tüm katılımcılara B-mod ultrason, sol ve sağ ortak karotid arterin uzak duvarında KİMK ölçümü yapılarak kaydedildi. Tüm grupların KİMK değerleri biyokimyasal, hormonal ve antropometrik değerlerle karşılaştırıldı. Verilerin normalliği Shapiro-Wilk testi ile test edildi. Mann-Whitney testi, Kruskal-Wallis ANOVA testi ve ki-kare testi uygulandı. Bulgular PKOS olan obez ve PKOS obez kadınların BMI, bel çevresi ve trigliserit değerleri normal ve PKOS kadınlara göre anlamlı derecede yüksekti. PKOS ve obez kadınlarda KİMK diğer gruplardakinden anlamlı olarak yüksekti (p<0.05); ancak diğer gruplar arasında istatistiksel olarak anlamlı bir fark yoktu (p>0.05). Sonuç PKOS hastalarında vücut kitle indeksi arttıkça KİMK artacağı, bu durumun ciddi kardiyovasküler sonuçlar doğurabileceği unutulmamalıdır. PKOS hastalarında obesite mevcutsa rutin takiplerde karotid arter ultrasonun yapılarak hayati risk oluşturabilecek kardiyovasküler hastalıkların erkenden önüne geçilebileceğini düşünmekteyiz.Öğe THE CYTOTOXIC EFFECTIVENESS OF PROPOLIS SAMPLES FROM SIVAS PROVINCE ON SK-OV-3 CELL LINE(17.09.2022) İrkin Sena Nur; Yurtcu, Nazan; Nas, Adalet; Durna Daştan, Sevgi; Güzel, Mehmet HakanHoney, pollen, propolis and other bee products are consumed as food from ancient times and are also natural products that are used as a traditional medicine in the treatment of many diseases [1-5]. The widespread use of all bee products to strengthen the immune system is a very popular topic in recent times. Today, the use of bee products in medical applications, as a food supplement, beverage substances, as a source of antioxidant, and antiaging in the cosmetic industries are becoming extremely common. Studies in the literature have shown that there are significant differences between the contents and biological activities of bee propolis from different botanical origins and different regions [6]. Within the scope of this study, propolis samples were obtained from various localities of Sivas, where beekeeping is intense. Propolis samples were obtained from beekeepers in the 2021 harvest period, and extractions were carried out in the laboratory using ethyl alcohol and water solvents. The scientific studies on bee products of Sivas province are very limited. In this study, it was aimed to determine and compare the cytotoxic activity of propolis samples collected different area of Sivas province in SK-OV-3 (Human ovarian cancer) cell line using MTT cell proliferation kit. MTT results were determined spectrophotometrically and IC50 values were calculated with the help of numerical data obtained by GraphPad Prism Version 8.4.1 software program. It was observed that ethyl alcohol isolates of propolis samples showed good antiproliferative activity in SK-OV-3 cell line (IC50 < 100 μg/mL). It is the first time to test the efficacy of propolis samples from Sivas region on the SKOV-3 cell line. It is thought that this study will be a beginning in terms of the therapeutic use of bee propolis samples from Sivas province and the preparation of new effective supplementsÖğe Histopathological Evaluation of Endometrium Cancers(16/09/2021) Göç, Rümeysa; Yurtcu, NazanIntroduction: Endometrial cancer is the fourth most common cancer of the genital tract, while it is the fourth most common malignancy in women. The aim of this study is to share the surgical treatment and results of cases who applied to our gynecological oncology clinic due to cancer, which is so common. Materials and Methods: All patients diagnosed with endometrial cancer and treated surgically between 2014-2021 were examined in this study, which was retrospectively planned in the Department of Obstetrics and Gynecology at Sivas Cumhuriyet University Faculty of Medicine. The results regarding the clinical, surgical and histopathological features of the patients were shared. Eighteen patients with sarcoma as a result of pathology and 2 patients with a second primary tumor were not included in the study. Results: The mean age of the patients included in our study was 59.28±11,45433. TAH + BSO procedure was performed in 181 patients, VAH procedure in 1 patient, endoscopic curettage in 16 patients, and endoscopic biopsy procedures in 2 patients. According to their diagnosis, 99 of the patients were endometrioid carcinoma and 25 of them were endometrioid carcinoma with squamous differentiation. It was stated that 2 patients were adenocarcinoma but the type was not specified. It was observed that 30 of them were endometrioid type, 7 of them squamous type and 1 of them serous type. In addition, we have 2 patients diagnosed with serous carcinoma in 12 patients, mucinous carcinoma in 3 patients, clear cell carcinoma in 12 patients, mixed carcinoma in 6 patients and malignant epithelial tumors. When mixed tumor grading is included, the number of grade 1 103 cases, grade 2 cases is 33, and grade 3 is 34. According to the histopathological results, the depth of myometrium invasion was found to be 8.554954955±7.386300184 and the myometrial wall thickness was 17.71621622±6.829107092. Cervical extension was observed in 30 materials, while lymphovascular involvement was observed in 44 materials. While endometrial hyperplasia was detected in 98 patients, the material of 102 patients was evaluated as atrophic endometrium. Nabothian cysts were detected in 129 cases, squamous metaplasia in 87 materials, and chronic cervicitis in 132 cases. Myometrial involvement was 132, corpus albicans 136, right ovary 136, right tuba 137, left ovary 135, left tuba 136. Conclusion: Endometrial cancer is the most common gynecological cancer. Tumor size, depth of myometrial invasion and histopathological grade are used to evaluate the risk of the disease. The importance of the depth of invasion was observed in FIGO 2009 in terms of staging and treatment.Öğe Third trimester cesarean scar pregnancy: a case report at 31 weeks of gestation(26/04/2021) Yurtcu, NazanCesarean scar pregnancy (SSG) is a rare form of ectopic pregnancy that develops with the implantation of the gestational sac over the scar of the previous uterine incision. It has become more common with the increasing number of cesarean sections around the world. The development and widespread use of transvaginal ultrasonographic examinations allows us to identifySSG diagnoses at more and earlier weeks. SSG carries a high risk of morbidity due to uterine rupture and excessive bleeding, and timely treatment is very important. In present study, scar pregnancy of 31 gestational weeks wasended. The diagnosis of pregnancy of a 37-year-old mother was first made by ultrasonography in the first trimester, and the diagnosis was confirmed by magnetic resonance imaging in the second trimester of pregnancy. The patient diagnosed with scar pregnancy did not accept the termination procedure and did not come to the control examinations.The patient applied to the emergency department at 31 weeks of gestation with the complaint of vaginal bleeding and cramping abdominal pain. Upon detection of intrauterine fetal death and uterine rupture, the patient underwent emergency cesarean operation and then hysterectomy when the bleeding did not stop.
