Bu çalışmada Prostaglandin F2 a analoğu olan latanoprostun, tek doz topikal uygulamasının göziçi basını düşürücü etkisi plesebo kontrollü olarak araştırıldı. Çalışma kapsamına oküler hipertansif 13 hastanın, 17 gözü alındı. Hastaların bazal göziçi basınçları ölçüldükten sonra topikal tek doz latanoprost uygulandı ve bunu takiben 2.,4.,6.,8.,12.ve 24. saatlerde göziçi basıncı(GİB) değişimleri ve yan etkiler değerlendirildi. Bazal ortalama GİB 25.71±3.42mmHg, 2.saatte 25.47+3.16mmHg, 4.saatte 22.76+2.97mmHg, 6.saatte 21.06±2.95mmHg, 8.saatte 18.59±2.72mmHg, 12 saatte 18.47±1.87mmHg ve 24. Saatte 19.65+2.18 idi(p<0.001). GİB'ındaki azalma 4. saatten sonra hem bazale göre hem de plaseboya göre istatistiksel olarak anlamlı idi. GİB'daki maksimum azalma 12. Saatte(%28.12) olduğu saptandı. Lokal yan etki olarak 1 hastada konjonktival hiperemi saptandı. Sonuç olarak topikal tek doz latanoprostun GİB'nı düşürmede etkili olduğu ve glokom tedavisinde yeni bir seçenek olduğu vurgulandı.
A Single Dose of Latanoprost Effects on the Intraocular Pressure In this study, effect of single dose of latanoprost that is an analogue of prostaglandin F2 a on intraocular pressure(IOP) was evaluated in 17 eyes of 13 patients. After measuring the basal IOP, single dose of latanoprost was applied and than at 2nd, 4th, 6th, 8th, 12th and 24th hours lOPs were measured and side effects were observed. Mean IOP was 25.71±3.42 mmHg before latanoprost and at 2nd, 4th, 6th, 8th, 12th and 24th hours latanoprost mean lOPs were 25.47±3.16 mmHg, 22.76+2.97 mmHg, 21.06±2.95 mmHg, 18.59±2.72 mmHg, 18.47±1.87 mmHg and 19.65±2.18 mmHg respectively (p<0.001). After 4 hours, reduction in IOP was statistically significant according to basal mean IOP and placebo group. Maximum reduction (%28.12) in IOP was detected at 12 hours. The ocular side effect was observed in 1 patient as conjunctival hyperemia. As a result, a single dose of latanoprost was effective in reducing IOP and it would be a new alternative in glaucoma therapy.
