dc.description.abstractGlucantime leyişmaniyoz tedavisinde kullanılan bir ilaçtır. Bu çalışmada eritrosit laktat dehidrogenaz aktivitesi (LDH), methemoglobin (MtHb) ve indirgenmiş, glutatyon (GSH) düzeyleri üzerine glucantim'in in vitro etkileri araştırıldı. Normal kan LDH için Km değeri 0,045 mM ve Vmax değeri 1,900 nmol (dk mg protein)-1 olarak bulundu, 3.75, 7.50 ile 15.00 mg meglumin antimonat içeren glucantimin, LDH'm Km degerini değiştirmezken (0,044, 0.043 ve 0,044 mM), Vmax degerini sırasıyla 0.675, 0.478 ve 0.324 nmol (dk mg protein)-1 olarak azalttigi bulundu. Eritrosit MtHb diizeyleri 0,15, 0.30 and 0.45 g meglumin antimonat içeren glucantim çözeltileri ile %0.33'ten %1.00, %1,62, ve %2.26 değerlerine yükseldi. Eritrositlerin GSH düzeylerinin 0.15, 0.30 and 0.45 g meglumin antimonat içeren glucantim çözeltileri ile 75 mg (dl eritrosit)-1 değerinden 57, 35 ve 12 mg (dl eritrosit)'-1 değerlerine azaldığı bulundu. Bu sonuçlar glucantim'in eritrosit LDH enzimini inhibe ettigini, MtHb duzeylerini arttırırken GSH düzeylerini azalttığını göstermiştir.en_US
dc.description.abstractGlucantime is used in the treatment of leishmaniasis. In this /study, the in vitro effects of glucantime on erythrocyte lactate dehydrogenase (LDH) activity, as well as the levels of methemoglobin (MtHb) and reduced glutathione (GSH) were investigated. The Km and Vmax values of erythrocyte LDH in the normal blood were found to be 0,045 mM and 1,900 nmol (min mg protein)-1 respectively. While the glucantime containing 3.75, 7.50 and 15.00 mg meglumine antimoniate did not change the Km values of LDH (0.044, 0.043 and 0.044 mM respectively), they increased the Vmax values to 0.675, 0.47S and 0.324 nmol (min mg protein)-1 respectively. The MtHb levels of erythrocytes were increased with glucantime solutions, containing 0.15, 0.30 and 0.45 g meglumine antimoniate, from 0.33% to 1.00%, 1.62%, and 2.26% respectively. The GSH levels of erythrocyte were decreased with glucantime solutions, containing 0.15, 0.30 and 0.45 g meglumine antimoniate from 75 mg (dl erythrocyte)-1 to 57, 35 and 12 mg (dl erythrocyte)-1 respectively. According to these results, it was established that glucantime inhibited erythrocyte LDH, increased MtHb levels and decreased GSH levels.en_US
