Öğe Association of plasma homocysteine level and carotid intima-media thickness in rheumatoid arthritis patients receiving methotrexate(Turkish League Against Rheumatism (TLAR), 2015) Hayta, Emrullah; Hizmetli, Sami; Atalar, Mehmet Haydar; Çinar, ZiynetObjectives: This study aims to evaluate the relationship between serum homocysteine levels and atherosclerotic plaques in carotid arteries in female patients with rheumatoid arthritis (RA). Patients and methods: Fifty-eight females with RA (mean age 45.50±11.69 years; range 42 to 60 years) and 22 female controls (mean age 47.3±8.9 years; range 38 to 62 years) with mechanical low back pain were included in the study. RA patients were divided into two groups: patients who were under methotrexate (MTX) therapy at least for two years (MTX group, n=32), and patients who did not receive MTX at least for past two years (non-MTX-disease-modifying antirheumatic drug group, n=26). RA patients’ disease activity scores in 28 joints were calculated. Carotid intima-media thickness and presence of atherosclerotic plaque were evaluated by high-resolution B-mode ultrasonography. Results: Plasma homocysteine levels were higher in MTX and non-MTX-disease-modifying antirheumatic drug groups compared to controls (16.88±6.84 and 10.37±2.54 ?mol/L, respectively) and the difference was statistically significant (p=0.001). In MTX, non-MTX-disease-modifying antirheumatic drug and control groups, atherosclerotic plaque was detected in 11 (34.5%) and four (15.4%) patients, and one patient (4.5%), respectively. Carotid intima-media thickness was significantly higher in MTX group compared to other two groups and the difference was statistically significant (p=0.002). Pearson correlation analysis revealed a significant correlation between intima-media thickness and homocysteine levels in MTX group (r=0.49, p=0.006). Conclusion: According to our study results, methotrexate treatment in female patients with RA increases plasma homocysteine levels and prevalence of atherosclerotic plaque. High homocysteine levels in these patients may be a cause of atherosclerosis. © 2015 Turkish League Against Rheumatism. Materials and Methods: This study was performed on 530 healthy adult males and females who have been living in central Sivas. The individuals evaluated as healthy using the applied questionnaire results were included in the study. Haematological parameters (eritrocyte, leukocyte, thrombocyte, mean thrombocyte volume, hemoglobin, hematocrit, mean corpuscular volume, mean corpuscular hemoglobin, mean corpuscular hemoglobin concentration and percentage neutrophil, lymphocyte, monocyte, eosinophil, basophil) were measured in the blood samples. Results: Normal values of studied haematological parameters of Sivas populace were determined by this study and the values obtained were evaluated according to sex and age groups. Conclusion: Obtained haematological values of adults who lived in Sivas were in accordance with the values reported as normal by other studies in the literature. Differences in some parameters could be originated from the methodology, geographical conditions, feeding habits of Sivas populace and the manner of lives.Öğe The investigation of some enzyme levels in the kidney, spleen, heart, liver and brain of Dami?nozi?de admi?ni?stered rats(University of Ankara, 2005) Göze, Ismihan; Yelkovan, Izzet; Bakir, Sevtap; Çinar, ZiynetIn this study, it was aimed to examine possible changes in enzyme activities and total protein levels in tissues of rats given daminozit at the dose of 150 mg/kg/day for 15. days introperitonly. Rats were killed by cervical dislocation on the end of 15 day. total protein (TP), alanin amino transferase (ALT), laktate dehidrogenase (LDH), alkaline fosfatase (ALP), gamma glutamil transpeptidaz (GGT), aspartat amino transaminaz (AST) enzyme activities were measured In prepared homogenates of spleen, kidney, heart, liver and brain tissues by Ciba Corning Express Plus autoanalyzer. GST enzyme was studied by spectrofotometric assay. The data of experimental group was compared with those of control rats. There were no statisticallyt differences between experimental group and control group in terms of enzymes in liver and kidney (p>0.05). There were significant differences in experimental group compared to control rats in spleen TP (u=32;p<0.05) and GGT (u-36;p<0.05) in heart GST (u=36;p<0.05) and brain GST (u=36;p<0.05) and GGT (u=32;p<0.05) enzymes (p<0.05), but the other enzyme activities did not show significant differences between two group (p>0.05).
