Öğe ASSOCIATION OF SERUM ANNEXIN A3 (ANXA3) LEVELS WITH AGE-RELATED MACULAR DEGENERATION Yaşa Bağlı Makula Dejenerasyonu İle Serum Anneksin A3 (Anxa3) Düzeylerinin İlişkisi(Yozgat Bozok Üniversitesi, 2023) Öztürk, Alpaslan; Gül, Mehmet Ali; Ağbektaş, Tuğba; Zontul, Cemile; Çiftçi, Cem Hakan; Taş, Ayça; Yeter, Duygu YalınbaşABSTRACT Objective: Age-related macular degeneration (AMD) is the most common cause of advanced visual loss in developed societies. ANXA3 contributes to tumor development by affecting cell signaling pathways, cell profiling, invasion, metastasis and cell apoptosis. The aim of this study was to compare serum ANXA3 levels in wet type and dry type macular degeneration patients with the control group and to investigate whether there is a significant difference between the groups. Material and Methods: A total of 78 patients (39 patients with wet AMD, 39 patients with dry AMD diagnosed at the same time period and 39 healthy controls without any previous diagnosis of macular degeneration) were enrolled in this study. ANXA3 levels were determined by enzyme-linked immunosorbent assay (ELISA). Results: Serum ANXA3 levels were found to be significantly higher in wet AMD than in the control group (p=0.005). In addition, the difference between serum ANXA3 levels of wet and dry AMD patients was found to be significant. It was observed that there was a significant increase in wet type AMD patients (p=0.004). However, no significant difference was found in ANXA3 levels in dry type AMD compared to the control group (p=0.444). Conclusion: These findings suggest that ANXA3 protein may play an important role in age-related macular degeneration.Öğe Diagnostic Significance of Chemerin and Chitinase 3-Like Protein 1 Levels in Cholelithiasis(2023) Öztürk, Alpaslan; Gül, Mehmet Ali; Zontul, Cemile; Güçlü, Gülşen Boztepe; Ağbektaş, Tuğba; Taş, Ayça; Topçu, ÖmerObjectives: Cholelithiasis is a common gallbladder disease that refers to the formation of gallstones. In this study, we investigated the relationship between Cholelithiasis and Chitinase 3-like-1 protein (Chi3L1) and Chemerin levels. Materials and Methods: Our study was conducted in 88 individuals, 44 of whom were healthy and 44 of whom had cholelithiasis. Chemerin and Chi3L1 serum levels were measured by enzyme-linked immunsorbent assay-ELISA method. The results were then analyzed using SPSS. Results: When cholelithiasis patient groups were compared with healthy groups, a significant increase was found in Chemerin levels in cholelithiasis patients (6.28-4.68, p<0.001). Chi3L1 concentration values were not statistically different between groups (p=0.460). After receiver operating characteristic analysis, the Chemerin result was statistically significant in predicting disease [p<0.001, area under the curve: 0.795 (0.697-0.893)], Chi3L1 was statistically insignificant in predicting disease. While the difference between the patient and control groups in terms of creatinine, albumin, amylase, alanine aminotransferase, aspartate aminotransferase, gamma glutamyltransferase, direct bilirubin, sodium, and calcium was statistically significant (p<0.05), the difference between Chi3L1 concentration and creatinine and total protein was statistically significant (p<0.05). Conclusion: Our study suggests that chemerin may be a new biomarker in the diagnosis of cholelithiasis. In addition, since our study has not been done before, it can bring a new perspective to the literature.Öğe Investigation of the relationship between ?2- adrenergic receptor (?2-AR) polymorphism and gastric cancer(2018) Atabey, Mustafa; Taş, Ayça; Ağbektaş, Tuğba; Bostancı, Meriç Emre; Topçu, Ömer; Siliğ, YavuzObjective: Gastric cancer is a multifunctional disease. Emotional stress, physiological and neuroendocrine changes incancer patients can lead to the activation of the hypothalamic-pituitary-adrenal axis and the release of hormonedependent hormones such as catecholamine. In particular, it has been reported that catecholamine induction directlyaffects the biological behavior of tumor cells via ?2-Adrenergic receptor (?2-AR) mediated signaling. In this study, itwas aimed to investigate polymorphism of ?2-AR gene (rs1042717) in gastric cancer patients.Method: Polymorphism in the ?2-AR gene (rs1042717) was determined by Real-Time PCR method in 60 gastric cancerpatients and 50 healthy controls. The results were evaluated using logistic regression and Chi-square (?2) test.Results: The comparison of gastric cancer patients and controls determined a statistically significant relationship foralcoholic drink consumption (p<0,05). There was a statistically significant relationship between ?2-AR (rs1042717)polymorphism and stomach cancer (p <0.05). There was a statistically significant relationship between mutant (AA)genotype with wild type (GG) and heterozygous (AG) polymorphic genotypes when evaluated in ?2-AR polymorphismgastric cancer patients and control group (?2: 19,38, p: 0.001). Similarly, there was a statistically significant correlationbetween heterozygous (AG) with wild type (GG) and mutant (AA) polymorphic genotypes in gastric cancer (?2: 14, 27,p: 0,001).Conclusions: In this study, it was found that the AG genotype is predominant in gastric cancer patients and controls,and that the AA genotype has a protective effect against gastric cancer.Öğe Investigation of the relationship between exportin5 (XPO5) polymorphism and gastric cancer(Sivas Cumhuriyet University, 2021) Tufa, Abdisa; Taş, Ayça; Ağbektaş, Tuğba; Topcu, Ömer; Genet, Solomon; Siliğ, YavuzObjective: We aimed to analyzed the association between gastric cancer and polymorphism of XPO5 gene.Method: The polymorphism in the XPO5 gene (rs11544382) was determined in 120 individuals (60 gastric cancer patients; 60 healthy controls) using Real-Time PCR method.Results: The comparison of gastric cancer patients and controls revealed a statistically significant relationship for alcoholic drink consumption (p<0.05).The relationship between XPO5 gene (rs11544382) polymorphism and gastric cancer was statistically not significant. There was no a statistically significant relationship between mutant (GG) genotype with both wild type (AA) and heterozygous (AG) polymorphic genotypes when evaluated in XPO5 polymorphism gastric cancer patients and control groups (χ2:0.12, p=0.729). The heterozygous (AG) was dominant in gastric cancers patients and control subjects, 93.3 and 91.7% respectively.Conclusions: This study provides information about alleleand genotype frequency distribution of XPO5 gene polymorphism (rs11544382) in Turkish At the same time, AG genotype was found to be dominant in gastric cancer patients and their controls.Öğe Mir146A polimorfizmi ile mide, kolon ve rektum kanserleri arasındaki ilişkinin incelenmesi(Cumhuriyet Üniversitesi, 2017) Ağbektaş, Tuğba; Siliğ, YavuzMikroRNA'lar yaklaşık 22 nükleotid uzunluğunda protein kodlamayan küçük RNA molekülleridir. MikroRNA ailesinin bir üyesi olan mir146a, hematopoetik hücrelerin denetlenmesi ve farklılaşması gibi birçok biyolojik yolağın düzenlenmesinde önemli bir rol oynamaktadır. Türk popülasyonunda, mir146a polimorfizmi ile mide, kolon ve rektum kanserleri arasındaki ilişki incelendi. Mir146a genindeki polimorfizim rs2961920 ve rs2910164 212 hasta (mide: 73, kolon: 76 ve rektum: 63) 77 sağlıklı kontrol de Real-Time PCR 'da allelik ayrımı belirlendi. Elde edilen sonuçlar, lojistik regresyon ve Khi-kare (?2) testi kullanılarak değerlendirildi. Mide, kolon, rektum kanserli hastalar ve kontroller de yaş, cinsiyet, sigara alışkanlığı ve ailede kanser hikayesi bulunması yönünden lojistik regresyon analizi ile değerlendirildiğinde, istatistiksel olarak anlamlı bir farklılık tespit edilmedi. Mide, kolon ve rektum kanseri hastalar ve kontroller alkol kullanma açısından değerlendirildiğinde, istatistiksel olarak anlamlı bir farklılık belirlendi (p<0,05). İncelenen Türk popülasyonunda mir146a rs2961920 polimorfizmi ile mide ve kolon kanserleri arasında istatistiksel olarak anlamlı bir farklılık bulundu (p<0,05). Fakat rektum kanseri arasında anlamlı bir farklılık tespit edilmedi (p>0,05). Mir146a rs2910164 polimorfizmi ile mide, kolon ve rektum kanserleri arasında istatistiksel olarak anlamlı bir farklılık saptanmadı (p>0,05). Ancak mir146a rs2910164 polimorfizmi mide kanseri hastaları ve kontrol grubunda değerlendirildiğinde, yabanıl tip (GG) ve heterozigot (CG) genotipleri biraraya getirilerek yapılan analizde bu polimorfizm ile mide kanseri arasında istatistiksel olarak anlamlı bir farklılık olduğu görüldü (?2:5,49, p:0,019). Benzer şekilde yabanıl tip (GG) ve homozigot (CC) genotipleri biraraya getirilerek yapılan analizde bu polimorfizm ile mide kanseri arasında istatistiksel olarak anlamlı bir farklılık olduğu saptandı (?2:5,39, p:0,020). The relationship between mir146a polymorphism and gastric, colon, rectum cancers have been investigated in Turkish population. Polymorphism in mir146a gene rs2961920 and rs2910164 have been determined in 212 patients (gastric: 73, colon:76 and rectum:63) and in 77 healthy controls by Real-Time PCR. Findings were evaluated by logistic regression and Khi (χ2) tests. The comparison of gastric, colon and rectum cancer patients and controls determined a statistically significant relationship for alcoholic drink consumption (p0.05). However, there was statistically significant relationship between this polymorphism and gastric cancer in GG+CG and CC genotypes when the gastric cancer patients and control group were evaluated for mir146a rs2910164 polymorphism (χ2: 5,49 p: 0,019). Similarly, there was statistically significant relationship between this polymorphism and gastric cancer in GG+CC and CG genotypes (χ2:5,39, p: 0,020). In this study, it is thought that by investigating the functions of microRNAs and their role in cancer, it may be promising in understanding the molecular pathology of cancer and in developing molecular targeted therapies.Öğe Radyoaktif iyot (I-131) tedavisi almış diferansiye tiroid kanseri hastalarında fox gen ailesi ile lncrna arasındaki ilişkinin araştırılması(Sivas Cumhuriyet Üniversitesi, 2023) Ağbektaş, Tuğba; Siliğ, YavuzTiroid kanseri, tüm kanserlerin yaklaşık %3'ünü oluşturmasına karşın endokrin kanserler içerisinde en sık görülen kanser türüdür. Son zamanlarda yapılan çalışmalarda mayadan insanlara kadar hemen hemen tüm ökaryotlarda bulunan çeşitli Forkhead box transkripsiyon faktörleri tanımlanmıştır. FOX genleri ile LncRNA'lar çeşitli kanser türlerinde doğrulandığı gibi, tümöral bölgelerin hücre çekirdeğinde spesifik olarak bulunmaktadır ve bu durum gen transkripsiyonunu düzenleyebileceğini göstermektedir. Bu çalışmada RAI tedavisi öncesi ve RAI tedavi sonrası diferansiye tiroid kanseri hastalarında FOX (FOXO1, FOXO3, FOXP1, FOXP2) ve LncRNA (MEG3, KCNQ1OT1, CASC21, HOXA11-AS) genlerinin ekspresyon düzeylerini RT-PCR ile ve protein düzeylerini ise ELİSA yöntemleri ile belirledik. RT-PCR analiz sonuçlarımızda, FOXO1 ve HOXO11-AS gen ekspresyon düzeylerinde RAI tedavisi öncesi hasta gruplarında anlamlı artış saptanırken, RAI tedavisi sonrasında bu genlerin ekspresyon düzeylerinde anlamlı bir azalış tespit ettik (p<0.05). FOXO3, FOXP1 ve FOXP2 gen ekspresyon düzeylerinde RAI tedavisi öncesi hasta gruplarında anlamlı artış, CASC21 gen ekspresyonunda anlamlı bir azalış saptadık (p<0.05). MEG3 ve KCNQ1OT1 gen ekspresyon düzeylerinde ise RAI tedavisi sonrası hasta gruplarında anlamlı bir artış gözlemledik (p<0.05). Ayrıca diferansiye tiroid kanserli hastalarda FOXO3 ve FOXP2 genleri ile MEG3, KCNQ1OT1, CASC21, HOXA11-AS genleri arasındaki korelasyon önemli bulundu (p<0.05). ELİSA yöntemi ile belirlediğimiz sonuçlarda ise, FOXP1 protein düzeylerinde anlamlı bir farklılık tespit edildi (p<0.05). Son olarak diferansiye tiroid kanseri hastalarının ameliyat sonrası, RAI tedavisi sırası ve RAI tedavisi sonrasındaki TSH ve Tg düzeyleri arasındaki fark anlamlı bulundu (x2=26.94, p<0.05).
