Öğe A comparative analysis of the fatty acid profiles in the liver and muscles of male and female Salmo trutta macrostigma(ELSEVIER SCI LTD, 2009) Akpinar, Mehmet Ali; Gorgun, Salih; Akpinar, Ali EmreThe fatty acid compositions of liver and muscle of male and female Salmo trutta macrostigma, in the Tohma River, Turkey, were determined by gas chromatography. There were quantitative differences between individual fatty acids in the tissues investigated, depending on the sex. The most abundant fatty acids in both tissues of both sexes were palmitic acid (C16:0; 19.0-21.6%), stearic acid (C18:0; 5.32-11.3%), C18:1 omega-7 (5.65-9.38%), oleic acid (C18:1 omega-9; 15.6-22.4%), eicosapentaenoic acid (EPA; C20:5 omega-3; 6.34-7.88%) and docosahexaenoic acid (DHA; C22:6 omega-3; 7.38-15.6%). The omega-3/omega-6 ratio in tissues were found to be 2.89 (male) and 1.97 (female) in liver, and 2.59 (male) and 2.26 (female) in muscle. S. trutta macrostigma may be a valuable food for human consumption in terms of fatty acids. (C) 2008 Elsevier Ltd. C16:0, C16: 1 n-7, C18:1 n-7, and C18:1 n-9 were the most abundant fatty acids in total and neutral lipids in all tissues investigated. In addition to these acids, C20:4 n-6 (arachidonic acid) and C22:6 n-3 (docosahexaenoic acid) were the other fatty acids having high percentages in the polar lipid fraction of the tissues. The highest and lowest n-3 to n-6 ratios, which are a good indicator of the quality of fish oils, were determined in the muscle (1.68) and liver (0.68) in the polar lipid fraction, respectively. It was determined that the species had low levels of the n-3 forms of polyunsaturated fatty acids in the investigated tissues. For a better understanding of fatty acid metabolism of V. vimba, more detailed studies are required in terms of feeding experiments.Öğe Effect of season on the fatty acid composition of the liver and muscle of Alburnus chalcoides (Guldenstadt, 1772) from Todurge Lake (Sivas, Turkey)(TUBITAK SCIENTIFIC & TECHNICAL RESEARCH COUNCIL TURKEY, 2012) Gorgun, Salih; Akpinar, Mehmet AliSeasonal variations in the liver and muscle fatty acid composition of Alburnus chalcoides (Guldenstadt, 1772) from Todurge Lake (Sivas, Turkey) were determined. The most abundant fatty acids in the investigated seasons and tissues were C16:0, C18:0, C18:1 n-9, C18:1 n-7, C20:4 n-6, C20:5 n-3, and C22:6 n-3. The total saturated fatty acids (SFAs) of the liver and muscle were at the highest in the spring (38.59%) and summer (41.65%), respectively. The total monounsaturated fatty acids (MUFAs) were at the highest in the autumn (51.79%) for liver and in the winter (40.04%) for muscle. The total polyunsaturated fatty acids (PUFAs) were determined to be at the lowest levels in the autumn for the both liver and muscle. The lowest total n-6 PUFA levels were determined as 2.60% and 4.19% in the autumn for the liver and muscle, respectively, while the lowest total n-3 PUFA levels were 14.84% for the liver (autumn) and 22.60% for muscle (spring).Öğe Fatty Acid Composition and omega 3/omega 6 Ratios in the Muscle of Wild and Reared Oncorhynchus mykiss(SPRINGER, 2015) Akpinar, Nukhet; Akpinar, Mehmet Ali; Gorgun, Salih; Akpinar, Ali EmreThe fatty acid composition and omega 3/omega 6 ratios in the muscle of wild and reared Oncorhynchus mykiss (rainbow trout) in the Tohma River (Sivas, Turkey) were determined by gas chromatography. There were quantitative differences between some fatty acids in the muscles of wild and reared rainbow trout. The most abundant fatty acids in both wild and reared fish were palmitic acid (16:0), stearic acid (18:0), oleic acid (18:1 omega 9), and docosahexaenoic acid (22:6 omega 3). The levels of omega 3 series and omega 6 series fatty acids in wild fish muscle were higher than in the reared fish. The omega 3/omega 6 ratios in muscle were found to be 1.92 (reared fish) and 2.28 (wild fish). The higher omega 3/omega 6 ratio in the wild fish might be a result of the season and the richer feeding conditions in the river under natural conditions. However, daily and continuously reared fish might be turned into more beneficial food for human consumption.Öğe Fatty Acid Composition of Diplolepis fructuum (Rubsaamen, 1895) (Hymenoptera: Cynipidae) During Its Developmental Stages(GAZI ENTOMOLOGICAL RESEARCH SOC, 2017) Akpinar, Mehmet Ali; Gorgun, Salih; Gencer, Lutfiye; Aktumsek, AbdurrahmanIn this study, fatty acid composition in lipid classes of Diplolepis fructuum (Rubsaannen, 1895) (Hymenoptera: Cynipidae), an insect species inducing gall formation on the rose hips (the genus Rosa), during its various developmental stages was analyzed. According to the results obtained, C 16:0, C 18:0, C 18:1 n-9, C 18:2 n-6 and C 18:3 n-3 were the most abundant fatty acids in lipid classes during developmental stages. In D. fructuum, the presence of C 20 polyunsaturated fatty acids could not be determined. The most important finding for the fatty acid analyses of the galls and the healthy fruits of the rose hips was that polyunsaturated fatty acid (PUFA) levels were higher in the healthy fruits.Öğe Fatty acid composition of Oncorhynchus mykiss during embryogenesis and other developmental stages(SPRINGER, 2006) Zengin, Hatayi; Akpinar, Mehmet AliFatty acid compositions of rainbow trout Oncorhynchus mykiss (Walbaum, 1792) was determined during embryogenesis, yolk-sac fry, swim up fry and fry stages. Embryonic and yolk-sac development of fertilized eggs were followed until fry weighed 25 g and samples were taken from each stage to analyse fatty acid composition. Analyses of fatty acid composition were performed by gas chromatography mass spectrometry. Throughout all the developmental stages of O. mykiss important changes, such as an increase in the percentage of 18:2, 18:3 and 20:5 and as a, decrease in the percentage of 14:0, 16:2, 20:0, 20:2, 20:3, 20:4 and 22:0 were observed in the amounts of fatty acid of developing eggs. During transition from the embryonic to the yolk-sac stage, an important increase in 20:0, 20:3, 20:5, 22:0 and a decrease in 18:3, 22:1 was determined between 45-day old embryo and yolk-sac fry. During transition from the 43 day yolk-sac stage to swim up stage, a decrease in the percentage of 18:0, 20:2 and an increase in the percentage of 14:0, 16:2, 18:2, 18:3 was seen. In all the observed stages, no qualitative but significant quantitative variations were determined in the fatty acid composition of O. mykiss. It may be concluded that 14:0, 16:0, 16:1, 18:0, 18:1 18, 20 and 22 polyunsaturated fatty acids are necessary for normal development of O. mykiss during their life stages.Öğe Fatty acid composition of the seeds of some species of Peucedanum (kral otu) from Turkey(Scientific and Technical Research Council of Turkey, 2012) Akpinar, Nükhet; Görgün, Salih; Akpulat, Hüseyin Aşkin; Akpinar, Mehmet Ali; Aktümsek, Abdurrahman; Akpinar, Ali EmreThe fatty acid compositions of the seeds of Peucedanum ruthenicum, Peucedanum chryseum, Peucedanum palimbioides, Peucedanum obtusifolium, and Peucedanum zedelmeierianum were investigated by gas chromatography (GC). The main fatty acids in the species were found to be palmitic acid (C 16:0, 5.78%-14.68%), oleic acid (C 18:1 ?-9, 31.28%-68.06%), and linoleic acid (C 18:2 ?-6, 15.99%-33.74%). Trace amounts of cis-vaccenic acid (C 18:1 ?-7, 0.82%-1.40%) were found in the seed oils of all of the species investigated. © TÜBITAK.Öğe Liver and muscle fatty acid composition of mature and immature rainbow trout (Oncorhynchus mykiss) fed two different diets(VERSITA, 2007) Gorgun, Salih; Akpinar, Mehmet AliFatty acid composition of liver and muscle tissues of immature and mature Oncorhynchus mykiss fed on two different diets were determined. Fatty acid analyses were carried out by gas chromatography. Palmitic acid (C16:0), oleic acid (C18:1 n-9), linoleic acid (C18:2 n-6) and docosahexaenoic acid (C22:6 n-3) were the major components in both liver and muscle tissues of immature and mature rainbow trout of both sexes. The amounts of C22:6 n-3 were higher in the liver (29.04 +/- 0.06 - 27.41 +/- 0.17%) and muscle (13.05 +/- 0.40 - 11.37 +/- 0.21%) of immature fish than in mature fish and depended on the composition of the diet. Results of this study show that fatty acid composition in fish tissues can considerably vary, depending on the age of fish and their diet. Thus more detailed studies are necessary on the influence of diet on immature and mature fish fatty acid composition. The age and diet of fish consumed may also be important for human health.Öğe Purification and Characterization of Lipase from the Liver of Carp, Cyprinus carpio L. (1758), Living in Lake Todurge (Sivas, Turkiye)(CENTRAL FISHERIES RESEARCH INST, 2012) Gorgun, Salih; Akpinar, Mehmet AliIn this study, a lipase from the liver of Cyprinus carpi Linnaeus (1758) living in Todurge Lake (Sivas) was purified with purification parameters of 90.38 mu mol/dk.mg protein specific activity and 75.50 fold purity. The purification procedure consisted of preparation of homogenate, precipitation with polyethylene glycol-6000 (PEG-6000) and chromatographic techniques including in Q sepharose, sephacryl S 200 HR and phenyl sepharose CL-4B, respectively. The purified lipase showed a single band with approximately a molecular weight of 74 kDa by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). pH and temperature optimums for the purified enzyme were determined as 8 and 37 degrees C using p-nitrophenyl butirate (p-NPB) as substrate. K-m and V-max values were found as 0.17 mM p-NPB and 2.6 mu mol/ml.dk, respectively.
