    Evaluation of antioxidant and anticancer effects of Thymbra sintenisii subsp. isaurica extract

    Background: The purpose of this study was to investigate the phenolic composition and antioxidant activity of Thymbra sintenisii subsp. isaurica extract (TSIE) and, to evaluate, for the first time, anticancer effect on human MCF-7 (breast carcinoma) cells. Materials and Methods: The antioxidant capacity of TSIE was evaluated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and total polyphenol assays. The anticancer activities of TSIE were tested on MCF-7 (breast carcinoma) cells. Results: Total polyphenol value of extracts TSIE was found as 73.02 mg gallic acid /g powder. DPPH result of IC50 value of TSIE was found to be 27.15 mu g/mL. To examine anticancer effect of TSIE at different concentrations were given on MCF-7 cells. TSIE was observed to reduce the cell viability in a dose-dependent manner. This anticancer property of the TSIE provides a highlights the importance of plant research for drug design. Conclusion: In this study, anticancer effects and antioxidant level of endemic species, that is TSIE, are evaluated on MCF-7 cells. Thus, an effective therapeutic agent for cancer treatment is aimed to develop. Further studies are needed to better understand the molecular mechanisms underlying this effect of TSIE.

    In vitro antimicrobial, antioxidant, cytotoxic activities, and wound healing potential of Thymbra capitata ethanolic extract

    Objectives: In this study, we aimed to detect the chemical compounds of Thymbra capitata ethanolic extract (TC-EtOH) as well as to evaluated its antimicrobial, antioxidant, cytotoxic activities, and in vivo wound healing effects. Methods: The chemical composition of TC-EtOH was analyzed by Gas chromatography-mass spectrometry (GC-MS). Antioxidant and antimicrobial properties were determined with 2,2-diphenyl-1-picrylhydrazyl (DPPH), disc diffusion test and broth micro-dilution (minimal inhibitory concentration [MIC]) methods, respectively. Cytotoxic activity was tested on MG63 (human osteosarcoma) and MCF-7 (human breast carcinoma) cells by 2,3-bis-(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxanilide (XTT) assay. Tumor necrosis factor alpha (TNF-alpha) protein levels were determined by ELISA. Results: The major components of TC-EtOH were tetratria contane (14.92%), camphor (12.50%), and terpineol (10.77%). TC-EtOH showed powerful antimicrobial activity in C. Tropicalis (0.03 mg/mL). The IC50 values of the TC-EtOH of the DPPH were determined 21.5 mu g/mL. The IC50 values were calculated 37.28 and 44.40 mu g/mL on the MG63 and MCF-7 cell lines, respectively. It was observed that the wounds treated with TC-EtOH showed a faster healing. Conclusions: According to results, T. capitata species are thought to be natural antioxidants and a novel pharmaceutical compound for the pharmaceutical industry.

    Protective Effect of Salvia cadmica on Fibroblast Cells from t-bhp-induced Oxidative Damage

    Background: Salvia species known as Sage are among the important aromatic plants used in the world. This study, it was investigated the antioxidant capacity of Salvia cadmica and to investigate its protective effect on oxidative damage in t-BHP-induced fibroblast cells. Methods: Antioxidant activity and phenolic characterization of the extract were evaluated using DPPH, TPC, TFC, FRAP, and HPLC, respectively. TAS, TOS, MDA and 8-oxo-guanine, CAT, SOD, and GPx values were examined using enzyme-linked immunosorbent assay (ELISA). The antiproliferative and apoptosis effects of Salvia cadmica ethanolic extract were determined using XTT assay and fluorescent probes in fibroblast cells. Results: As a result of GC-MS analysis of Salvia cadmica ethanolic extract, carvacrol content was found to be high. The IC(50 )value of the DPPH antioxidant assay of the Salvia cadmica ethanolic extract was 80 +/- 0.51 mu g/mL. TPC, TFC, and FRAP values were found to be 18.25 +/- 0.64 (mg gallic acid/g powder), 1.691 +/- 0.314 (mg quercetin /g powder), 31.5 +/- 0.10 (mg Trolox/g powder), respectively. Total antioxidant and TOS values were found to be 0.383 +/- 0.033 (mmol Trolox Equ L-1), 16.31 +/- 0.71 (mu mol H2O2 L-1) for 0.25 mg/mL, and 0.725 +/- 0.05 (mmol Trolox Equ L-1), 12.02 +/- 0.56 (mu mol H2O2 L-1) for 0.5 mg/mL. In addition, while CAT and GPx significantly decreased enzyme activities, no significant change was observed in SOD enzyme activity. Ethanolic Salvia cadmica extract exhibited apoptotic features compared to only the t-BHP group. Conclusion: These results suggest that Slavica cadmica extract works through a free radical mechanism.
