Öğe Evaluation of 4-Hydroxy-2-Nonenal, Dityrosine and 8- Hydroxy-2-Deoxyguanosine Expressions in Lambs withWhite Muscle Disease(2021) Karakurt, Emin; Karataş, Özhan; Dag, Serpil; Beytut, Enver; Mendil, Ali Sefa; Nuhoğlu, Hilmi; Yıldız, AyferThe aim of this study was to investigate Reactive Oxygen Species (ROS)-induced lipidperoxidation, protein and DNA damage with oxidative stress markers such as 4-hydroxy-2- nonenal (4-HNE), dityrosine (DT) and 8-hydroxy-2-deoxyguanosine (8-OHdG) in lambs withWhite Muscle Disease (WMD). The material of this study consisted of tissue samples taken from20 sheep with WMD and 6 healthy control groups brought to Pathology Department for routinehistopathological diagnosis between 2012 and 2019. In macroscopic examinations, chalky-whitelarge necrotic areas on the epicardial and especially on the endocardial surfaces of the right andleft ventricular walls were observed. In microscopic examinations, it was observed that thedegenerated muscle fibers in the heart were homogeneously pink in color, lost their cytoplasmiccitration, swelled and their nuclei were pyknotic. Calcifications in necrotic and degenerated areaswere detected. All white muscle disease cases were immune positive for 4-HNE, DT and 8-OHdGexpressions in heart tissues. A significant difference (P<0.05) was detected between the groups interms of immunoreactivity to 4-HNE, DT and 8-OHdG in immunohistochemical andimmunofluorescence staining findings. In conclusion, our study showed that formation of ROS thatis important in pathogenesis of WMD causes not only lipid peroxidation, but also proteinmodification and DNA damage.Öğe Expression of PCNA, MMP-9, p53, Bax and Bcl-2 in canine transmissible venereal tumors(İstanbul Veteriner Hekimler Odası, 2020) Karakurt, Emin; Kuru, Mushap; Dağ, Serpil; Beytut, Enver; Nuhoğlu, Hilmi; Demir, Murat Can; Kaçar, CihanIn this study, we aimed to evaluate the proliferative, metastatic and apoptotic capacities of TVT cases, which are in various phases of development, by using immunohistochemical markers. The material of this study consisted of twelve female and six male dogs diagnosed with TVT brought to our department between 2007 and 2020 years. Diff-quick staining was applied to the smear taken from tumoral masses for cytological examinations. Tumoral tissues from dogs were fixed in a 10% neutral buffered formaldehyde solution. After routine tissue procedures Hematoxylin & Eosin stain was applied to the sections. Tissue sections were investigated under a light microscope and photographed. Immunohistochemical staining was performed on the tissues using the avidin-biotin immune peroxidase complex method. As a result of macroscopic, cytological and histopathological examinations, TVT positive cases were divided into three according to their developmental stages. While the expression of PCNA, MMP-9, mutant p53 and Bcl-2 increased significantly in progressive cases compared to regressive and stable cases, Bax expression increased significantly in regressive cases compared to progressive and stable cases. In conclusion, we thought that the mentioned markers are very useful for understanding the prognosis of TVT, the tumor aggressiveness and the survival of the malignant cells.Öğe Investigation of ruminant encephalitic and septicemic listeriosis by the immunofluorescence method(2021) Karakurt, Emin; Büyük, Fatih; Dağ, Serpil; Beytut, Enver; Çelebi, Özgür; Karataş, Özhan; Nuhoğlu, HilmiListeria genus is Gram-positive facultative intracellular bacteria that are found in the soil as saprophytesbut also contain species that have pathogenic effects in their last host. Listeria monocytogenes is the major pathogenin Listeria species that is responsible for the majority of Listeriosis cases in humans and animals. In this study, itwas aimed to investigate L. monocytogenes with histopathological and immunofluorescence (IF) methods in brainand liver tissues taken from sheep and cattle with clinical signs of suspicious Listeriosis. In the study, isolation andidentification of L. monocytogenes with cultural methods were also applied. The material of the study consistedof tissue samples from 16 sheep and 2 cattle. As a result of cultural analysis, L. monocytogenes was identifiedfrom 12 (66.6%) of cases. Within the framework of histopathological and macroscopic findings, 16 (88.8%) of 18cases with suspected Listeriosis were positive with the immunofluorescence method. These findings reveal that theimmunofluorescent staining method is a reliable and sensitive method in the diagnosis of Listeriosis.
