Öğe Bir Buzağıda Çoklu Kongenital Anomali Olgusu(2020) Yurdakul, Ibrahim; Yalçın, Mustafa Onur; Karataş, ÖzhanBu olgu sunumunda Simmental ırkı 2 günlük bir buzağıda karşılaşılan koksigeal agenezi, atresia ani et vulva, vaginal ektopi ile beraber seyreden çoklu kongenital anomali olgusunun tanısı ve sağaltım sonuçları değerlendirildi. Olguyu kuyruk, anüs ve vulvanın olmaması şikayeti ile kliniğimize getirilen 2 günlük Simmental ırkı bir buzağı oluşturdu. Yapılan klinik muayenede; kuyruk agenezisi, atresia ani et vulva ve buzağının dışkı ve idrarını perineal bölgede ektopik vaginadan köken alan yumuşak fluktuan bir kitlenin delinmesi sonucu yapmakta olduğu saptandı. Operatif olarak uzak-laştırılan kitlenin histopatolojik incelemesinde vagina mukozası ile uyumlu non-keratinize çok katlı yassı epitel belirlen-di. Sonuç olarak; nadir olarak rastlanılan bu vakanın klinik pratiğe ve literatüre katkı sağlayacak nitelikte olduğu düşünüldü.Öğe Determination of local expressions of IGF-1, LC3B and NF-kB in white muscle disease in lambs by immunohistochemical method(2024) Akçakavak, Gökhan; Karataş, Özhan; Cifci, Aysenur Tural; Dağar, Osman; Doğan, Osman; Tuzcu, MehmetWhite muscle disease (WMD) is also known as Stiff Lamb Disease or Nutritional Muscular Dystrophy. Selenium and/or Vitamin E deficiency constitutes the etiology of the disease. This study aimed to immunohistochemically evaluate local protein expressions of Nuclear factor kappa B (NF-kB), Insulin-like growth factor-1 (IGF-1) and Microtubule-related protein 1A/1B-light chain 3 beta (LC3B) in WMD. The material of the study consisted of 15 WMD, and 6 healthy lamb heart samples. The heart tissues of the autopsied lambs were subjected to routine tissue processing and paraffin blocks were obtained. Then, it was stained with Hematoxylin-Eosin and immunohistochemical methods. Control group lambs had normal macroscopic appearance. Macroscopically, hyaline degeneration and zenker’s necrosis, calcification areas were observed in WMD tissues. Microscopically, degenerative and necrotic muscle fibers, calcification areas, fibrosis, mononuclear cell infiltrates and macrophage infiltrates were detected in WMD heart tissues. Immunohistochemically, significant increases were detected in IGF-1 (p<0.001), LC3B (p<0.001) and NF-kB (p<0.05) in the WMD group compared to the control group. Immunoreactivity in the relevant primers was detected commonly in degenerative and necrotic muscle fibers. In addition, occasional immunoreactivity was observed in the relevant primers in inflammatory cell infiltrates. In conclusion, NF-kB, IGF-1 and LC3B protein expressions were evaluated immunohistochemically for the first time in lambs with WMD. Our findings show that IGF-1 and LC3B proteins are highly expressed in heart tissue in WMD. Additionally, it is possible to say that IGF-1 and LC3B can be used in the diagnosis of WMD.Öğe Determination of the Effects of Helichrysum Plicatum DC. Subsp. For this purpose, a total of 24 Sprague Dawley rats, 6 each in four groups, were used in the study. Rats were administered a single dose of TS via IP and HPE via oral gavage for 8 days. After the administrations, the rats were sacrificed and blood and testicular tissues were collected. Testicular tissues were preserved for use in biochemical, histopathological, immunohistochemistry and in situ hybridization analyzes. The cauda epididymis was trimmed by separating from the testis and the resulting liquid was used for semen analysis. The sperm motility decreased and the rate of dead and abnormal spermatozoa increased in parallel to the increase (PÖğe Effects of Rhodiola rosea on indomethacin-induced gastric injury(2021) Okkay, Irmak Ferah; Okkay, Ufuk; Karataş, ÖzhanObjective: The objective of the present study was to evaluate the effects of Rhodiola rosea in the indomethacin-induced ulcer model in rats and to clarify the underlying mechanisms of action. Methods: Rats in treatment groups were treated with Rhodiola rosea (RR) 14 days. Peptic ulcer was induced by indomethacin (IND) injection (100 mg/kg, p.o.). The groups (n = 6) were designed as; Group I (control); Group II (IND): After 24h of food starvation, rats were given only 100 mg/kg IND by oral gavage to induce gastric mucosal injury. Group III (ESO): Rats were pretreated with 20 mg/kg of ESO for 14 consecutive days by oral gavage. Group IV (RR): Rats were pretreated with 500 mg/kg RR for 14 consecutive days with oral gavage. Results: Rhodiola rosea effectively alleviated indomethacin-induced ulcer via reduction in oxidative stress (decreased MDA and increased SOD, and GSH). Moreover, Rhodiola rosea alleviated indomethacin-induced damage by regulating expressions of COX enzymes, prostaglandin E2, proliferating cell nuclear antigen (PCNA), cell proliferation, apoptosis and regulated the NF-?B signaling pathway. Rhodiola rosea also attenuated inflammatory injury by suppressing TNF-????, IL-1?, and NF-?B. The caspase-3 expression was also down-regulated in stomach tissues. Conclusions: In conclusion, Rhodiola rosea protected the gastric mucosa from harmful effects of indomethacin and as a natural medicinal herb, Rhodiola rosea might be a potential therapeutic agent for preventing and treating indomethacin-induced gastric damage.Öğe Evaluation of 4-Hydroxy-2-Nonenal, Dityrosine and 8- Hydroxy-2-Deoxyguanosine Expressions in Lambs withWhite Muscle Disease(2021) Karakurt, Emin; Karataş, Özhan; Dag, Serpil; Beytut, Enver; Mendil, Ali Sefa; Nuhoğlu, Hilmi; Yıldız, AyferThe aim of this study was to investigate Reactive Oxygen Species (ROS)-induced lipidperoxidation, protein and DNA damage with oxidative stress markers such as 4-hydroxy-2- nonenal (4-HNE), dityrosine (DT) and 8-hydroxy-2-deoxyguanosine (8-OHdG) in lambs withWhite Muscle Disease (WMD). The material of this study consisted of tissue samples taken from20 sheep with WMD and 6 healthy control groups brought to Pathology Department for routinehistopathological diagnosis between 2012 and 2019. In macroscopic examinations, chalky-whitelarge necrotic areas on the epicardial and especially on the endocardial surfaces of the right andleft ventricular walls were observed. In microscopic examinations, it was observed that thedegenerated muscle fibers in the heart were homogeneously pink in color, lost their cytoplasmiccitration, swelled and their nuclei were pyknotic. Calcifications in necrotic and degenerated areaswere detected. All white muscle disease cases were immune positive for 4-HNE, DT and 8-OHdGexpressions in heart tissues. A significant difference (P<0.05) was detected between the groups interms of immunoreactivity to 4-HNE, DT and 8-OHdG in immunohistochemical andimmunofluorescence staining findings. In conclusion, our study showed that formation of ROS thatis important in pathogenesis of WMD causes not only lipid peroxidation, but also proteinmodification and DNA damage.Öğe Evaluation of a sternum dehiscence reconstruction graft on an animal model(2022) Kasımzade, Ferit; Ada, Fatih; Karataş, ÖzhanSternum dehiscence occurring after surgeries requiring median incision of the sternum remains a current problem. New materials are needed to prevent or reduce the risk\rof developing sternum dehiscence. In this study, the effects of extracellular matrix (ECM)-like sternum dehiscence graft in a rabbit model were investigated. A total of\r24 rabbits in three groups were used in the study. There were eight rabbits in each group. The first group was the control group, the second group was the conventional\rclosure method, and the third group was the sternum closure graft group. Median sternotomy was applied to the rabbits in the second and third groups. Animals were\rsacrificed 4 weeks after surgery and sternum samples were taken. Sternum samples taken from untreated animals had normal histological appearance. Cartilage formation\rand fibrocyte-fibroblast activity were mild in the conventional surgery group, while moderate in the reconstruction graft group. It has been shown histopathologically that\rsternum closure graft accelerates bone healing (P= 0.015). It is obvious that if this graft is used clinically, it will reduce the incidence of sternum dehiscence.Öğe Evaluation of ultrasonic intravascular thrombectomy system on a rabbit model in the treatment of deep vein thrombosis(Selçuk BAŞAK, 2021) Kasimzade, Ferit; Ada, Fatih; Karataş, ÖzhanBackground/Aim: Advanced treatment options are needed in deep venous thrombosis (DVT), which is a special subgroup of venous disease. We examined this subgroup on an animal model and aimed to evaluate the effects of the ultrasonic intravascular thrombectomy system on vascular endothelial damage. Methods: A total of 24 rabbits in 3 groups were used in the study. DVT was created in the common iliac vein by the administration of intravascular fibrin. One hour passed for DVT formation. The ultrasonic intravascular thrombectomy system and the mechanical thrombectomy system were used separately in the DVT groups. After one hour, samples obtained from the groups were examined histologically. Listeria monocytogenes is the major pathogenin Listeria species that is responsible for the majority of Listeriosis cases in humans and animals. In this study, itwas aimed to investigate L. monocytogenes with histopathological and immunofluorescence (IF) methods in brainand liver tissues taken from sheep and cattle with clinical signs of suspicious Listeriosis. In the study, isolation andidentification of L. monocytogenes with cultural methods were also applied. The material of the study consistedof tissue samples from 16 sheep and 2 cattle. As a result of cultural analysis, L. monocytogenes was identifiedfrom 12 (66.6%) of cases. Within the framework of histopathological and macroscopic findings, 16 (88.8%) of 18cases with suspected Listeriosis were positive with the immunofluorescence method. Endothelial damage remains a problem in the endovascular treatment of peripheral artery CTO. In our study, this subgroup was examined with an ultrasonic total occlusion system for vascular endothelial damage on an animal model. Methods: We used twenty-four rabbits divided equally into three groups, created chronic total occlusion in the common iliac artery by applying a bioabsorbable polymer sponge, and waited four weeks for CTO formation. After four weeks, the samples obtained from the groups were examined histologically. Ancak karaciğer ve böbrek fonksiyonları üzerine belirgin yan etkilerinin olması klinik yararlılığını sınırlamaktadır. Okaliptolün (OKA, 1.8-sineol) antioksidan ve antiinflamatuar tıbbi aktiviteleri farklı çalışmalarda bildirilmiştir. Bu çalışmada GM kaynaklı hepatotoksisite üzerine OKA’nın etkilerinin değerlendirilmesi amaçlanmıştır. Çalışma grupları kontrol (K), OKA, GM ve GM + OKA'dan oluşmaktaydı ve her grupta 7 rat bulunmaktaydı. Although CP has strong anticancer properties, its clinical use is limited due to side effects such as ototoxicity, neurotoxicity, myelosuppression and nephrotoxicity. Taxifolin (Tax) is reported to exhibit various possess effects such as anti-inflammatory, antioxidant, antimicrobial, antiviral and anticancer. In this study, we aimed to investigate the possible effects of Tax on CP-induced nephrotoxicity. This study consisted of Control (C), Taxifolin (Tax), Cisplatin (CP) and Cisplatin + Taxifolin (CP + Tax) groups, and there were 6 rats in each group. CP was administered to rats intraperitoneally (i.p.) in a single dose of 7 mg/kg, and Tax was administered orally at a dose of 50 mg/kg for 7 consecutive days. Histopathologically, significant changes such as tubular epithelial degeneration and necrosis, tubular dilatation, inflammatory cell infiltrates, hyaline cast, and glomerular atrophy were detected in the CP group.
