    MOTAKK, as a national external quality control program has been launched to evaluate the molecular detection of viral infections including HBV DNA and HCV RNA in molecular microbiology diagnostic laboratories in Turkey. This program is prepared in compliance with ISO 17043:2010 (Conformity assessment general requirements for proficiency testing) standards, and aims to take the place of external quality control programs from abroad, contributing to standardization and accuracy of molecular diagnostic tests in our country. The aim of this study was to evaluate 2015 and 2016 results of the MOTAKK External Quality Control Program for HBV DNA and HCV RNA viral load. The calls were announced on the web page of MOTAKK (www.motakk.org). The quality control samples were sent to participating laboratories in 2015 and 2016. Main stocks were prepared from patients with chronic hepatitis B and C who had viral load detection with reference methods according to WHO reference materials for viral load studies to improve quality control sera. From these main stocks, samples with different viral loads were prepared from dilutions of plasma with HBV, HCV, HAV, HIV, Parvovirus B19 and CMV negative serologic markers. Quality control samples were sent to the participating laboratories along with the negative samples in the cold chain. The laboratories accomplished the related tests within 2-3 weeks and entered their results on the MOTAKK web page. These results were analysed according to ISO 13528 (Statistical methods for use in proficiency testing by interlaboratory comparison) and scoring reports were created by a software developed by MOTAKK and sent to participating labs. Each laboratory evaluated their own results in comparison with the other laboratory results, reassessed the tests via observing the distance from the mean result and the reference values. The number of laboratories participating in the HBV DNA and HCV RNA external quality control program was 70-73 in 2015-2016. Participants were able to comply with the program tools, registering, entering results and receiving the results reports problem. In HBV panel, 72.6-89.1% and 84.7-90.3% of the participant laboratories were in 1 standard deviation (SD) in 2015-2016, respectively. In HCV panel, 70.8-89.1% and 84.7-90.3% of the participant laboratories were in 1 SD in 2015-2016, respectively. A national external quality control program for HBV DNA and HCV RNA in Turkey has been prepared for the first time with this project and implemented successfully. All the data provided in the MOTAKK external quality control program final report, compensate all the data provided by the quality control program final reports from abroad; additionally, the report allows comparison of used technologies and commercial products.

    Isolation and Antibiotic Resistance Rates of Mycobacterium tuberculosis complex in Mycolor TK® System

    Objective: Tuberculosis (TB) is a public health problem. This study aimed to determine the growth rates and drug susceptibility levels of patients with Mycobacterium tuberculosis complex (MTC) growth in cultures obtained and to compare the results with the growth rates and drug susceptibility levels found in our country and other countries. It also aimed to evaluate the results of supplementing classical methods such as Lowenstein-Jensen (LJ) with liquid TK MEDIUM (R) and to determine the relationship between the growth rates obtained with both methods. Materials and Methods: The study included patients who were Erlich Ziehl Neelsen (EZN) positive or negative and whose samples showed MTC growth in culture and susceptibility to the main drugs used in the treatment of TB, including streptomycin (SM), isoniazid (INH), rifampicin (RIF) and ethambutol (EMB). Patient samples were inoculated simultaneously into liquid TK (R) MEDIUM based on color change and LJ solid medium. TB identification and antimicrobial susceptibility testing were performed using the TK ANTI TB and PNB KIT (R) for the Mycolor TK (R) automated mycobacterial culture system. Results: The study found resistance to dual drugs at 5.4%, INH+RIF at 1.35%, INH at 6.8%, RIF at 14.8%, EMB at 17.6%, and SM at 1.35%. Out of 69 samples cultured in LJ medium, three failed to grow in liquid TK (R) MEDIUM, and two of 71 samples cultured in liquid TK (R) MEDIUM gave negative results in solid LJ medium. Conclusion: We found that the drug resistance rates we obtained in our study were similar to the results of the Republic of T & uuml;rkiye Ministry of Health and other studies conducted in our country. In a joint study conducted with liquid and solid media, no difference was found in the detection of TB bacilli. Mycolor TK (R) automated system can enable rapid mycobacterial culture in laboratories with limited resources.

    Mortality rates and risk factors associated with mortality in patients with stenotrophomonas maltophilia primary Bacteraemia and Pneumonia

    This study aims to evaluate the risk factors associated with the mortality of S. maltophilia infections. Patients aged 18 years and older with S. maltophilia infection. Patients were divided into two groups primary bacteraemia and pneumonia. Of 176 S. maltophilia infections, 85 (48.2 %) were classified as bacteremia and 91 (51.8 %) as pneumonia. The mortality rate was 56 %, with no significant difference observed between the groups. Invasive mechanical ventilation, history of carbapenem use, and high Charlson Comorbidity Index (CCI) were significantly higher in the pneumonia group. In univariate analysis, higher Acute Physiology and Chronic Health Evaluation (APACHE) II score, higher Sequential Organ Failure Assessment (SOFA) score, and use of total parenteral nutrition (TPN) were identified as independent risk factors for 28-day mortality. This study demonstrates a mortality rate of 56 % in S. maltophilia infections and provides concrete data on risk factors for mortality.

    The frequency of intestinal parasites in primary school children in urban and rural regions

    The aim of this study was to determine the prevalence of intestinal parasites in primary school children in Sivas and to consider the changes in frequency and variety of intestinal parasites in the light of previous studies carried out since 1985. Specimens were examined by direct microscopy and staining. In addition, the relationship of intestinal parasites with physical development, success and socio-economic situation was investigated using surveys. Intestinal parasites were found in 191 (26.2%) of 730 stool specimens, 88 (12.9%) of cellophane tape specimens and parasites were found in both types of specimens obtained from 251 (37.1%) out of 677 students. The most common five parasite species were Giardia intestinalis (13.7%), Enterobius vermicularis (12.8%), Entamoeba coli (7.4%), Blastocystis hominis (2.2%) and Hymenolepis nana (1.2%). Intestinal parasites were most frequently detected in the Celtek Village Primary School (50.0%). This school was followed by Imranli YIBO School (45.7%), 60.Yil Primary School (36.3%) and Koyulhisar YIBO (21.3%) School. We found that there was no statistically important relationship between the occurrence of parasites with weight, length and success. It was related to the socioeconomic situation. Furthermore, in this region when the previous studies in similar age groups are considered, a decrease in intestinal parasites frequency and variety was revealed.
