Öğe Biological Activity Evaluation of Scorzonera tomentosa L.(Siirt Üniversitesi, Ağustos 2023) Ulger, DilaraAbstract: This study aimed to evaluate the phytochemical components, antimicrobial activity, and antioxidant activity of 80% ethanol extract of Scorzonera tomentosa, an endemic species. The chemical constituents of the ethanolic extract of S. tomentosa was primarily characterized by gas-chromatography-mass spectrometry analysis (GC-MS), ten components were identified. The major component was found as 2-pentanamine (35.68%). When the antioxidant capacity of S. tomentosa was examined, 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activities were determined to be quite high compared to the reference drug (IC50 values; DPPH: 517.0 ± 1.86 µg mL-1; ABTS: 244.8 ± 0.94 µg mL-1; reference drug: 1.313 ± 0.24 µg mL-1). In addition, according to total phenol content and total flavonoid content analyses, it was determined that the plant is richer in flavonoids. The antimicrobial activity of this species is not at an effective level. More extensive studies with S. tomentosa may allow the plant to be used as a natural antioxidantÖğe Levels of oxidative stress and apoptosis-related biomarkers in endometriosis(Cukurova Üniverstesi, 2023) Ülger Özbek, Dilara; Sevtap Bakir; savaş karakuşAbstract Öz Purpose: In this study, the apoptosis marker M30, the oxidative stress markers malondialdehyde, (MDA) and asymmetric dimethyl arginine (ADMA) have been studied in the context of endometriosis. Materials and Methods: This prospective case-control study comprises 31 patients diagnosed with endometriosis and 31 controls. ADMA and M30 levels in blood serum were measured by the enzyme-linked immunosorbent assay (ELISA) method, and MDA levels were measured by the spectrophotometric method. In addition, some biochemical parameters and cancer antigen-125 (CA-125) levels were also measured. Results: M30 levels were statistically lower in endometriosis patients (271.5 IU/L) than in controls (371.3 IU/L). ADMA levels were higher in endometriosis patients (19.3 ng/L) compared to controls (12.7 ng/L). CA-125 levels were statistically higher in the endometriosis patients (65.1 U/mL) compared to the controls (19.0 U/mL). There was no significant difference between the two groups in MDA levels. The results regarding dyspareunia, pelvic pain, AST, and ALP were statistically significant. Conclusion: In our study, decreased M30 levels in the patient group were associated with reduced apoptosis in endometriosis. ADMA levels, elevated with the increase of oxidative stress, were higher in the patients. MDA levels, an indicator of increased oxidative stress, were also higher in the patient group. This study constitutes the first data regarding endometriosis patients' ADMA, M30, and MDA levels.Öğe Agmatine, telomerase and trace minerals levels in patients with obstructive sleep apnea syndrome(Şubat 2023) Ülger Özbek, Dilara; Sevtap Bakir; ömer tamer doğanObstructive sleep apnea syndrome (OSAS) is a disease characterized by repetitive, partially or com plete upper airway obstructions resulting in hypoxia and bioelectrical wakefulness reactions along with sleep. There is a limited and different information on the biochemical pathways that may determine harmful effects in OSAS patients with different disease severity. Hence, we aimed to estimate the plasma levels of polyamine agmatine, which has many effects on the central nervous system, telomerase and trace minerals in patients with OSAS. The study included 90 volunteer patients diagnosed with OSAS and divided into three groups of 30 people each according to the apnea-hypopnea index (AHI) score: mild, moderate and severe. Nocturnal blood oxygen saturation percentage (SpO2 ) and body mass index (BMI) were measured. Plasma agmatine level was defined by ultra-high-pressure liquid chromatography (UHPLC), plasma trace elements (Cu, Co, Mg, Mo, Zn, Se) level by inductively coupled plasma mass spectrometer (ICP-MS) and serum telomerase level by enzyme-linked immunosorbent assay (EL ISA) method. It was found that SpO2 value decreased as the disease progressed and showed a negative correlation with BMI, Co and Se plasma levels. The levels of agmatine and telomerase were shown to lower in patients with severe OSAS group compared to other groupsÖğe DNA Barcoding of Commercial Cockroaches in Turkey(2023)Accurate species identification has become a precondition for accomplished biodiversity administration and further genetic research. Species acquaintance technics require molecular tools such as DNA barcoding as well as morphological identification for accurate identification. Particularly, the application of subunit I of the mitochondrial cytochrome c oxidase (COI) gene for DNA barcoding for insects has approved to be very useful in species acquaintance. The main aim of this study is to generate the first reference library of DNA barcode for cockroaches in Turkey using previously published data. As a result of the literature research, it has been observed that no study has been carried out on the DNA barcode of Turkish cockroaches. Therefore, in this study, we evaluated the advantage of DNA barcoding applied to two cockroach samples from Turkey for the first time. Our working samples implicated 10 DNA barcodes grounded on sequences created from our present study and 109 other DNA barcodes from BOLD. Various molecular analyzes including genetic distance-origin assessment (NeighborJoining and Maximum Likelihood trees) has been applied to accurately identify and describe species. In addition, Blaptica dubia (B. dubia) (Serville, 1838) and Nauphoeta cinerea (N. cinerea) (Olivier, 1789) have been reported as the first country records. It has been observed that reference libraries like BOLD are not yet sufficiently populated with COI sequences of Turkish cockroach species. In order for Turkish cockroach bio-assessment and biodiversity studies to benefit from the advantages of DNA barcoding, it is of great importance that cockroach inventories and taxonomic studies include DNA barcodes.Öğe Investigating the effect of ribavirin treatment on genetic mutations in Crimean-Congo haemorrhagic fever virus (CCHFV) through next-generation sequencing.(2023) Pektaş, Ayşe NurCrimean-Congo haemorrhagic fever (CCHF) is the most widespread tick-borne viral haemorrhagic fever affecting humans, and yet a licensed drug against the virus (CCHFV) is still not available. While several studies have suggested the efficacy of ribavirin against CCHFV, current literature remains inconclusive. In this study, we have utilised next-generation sequencing to investigate the mutagenic effect of ribavirin on the CCHFV genome during clinical disease. Samples collected from CCHF patients receiving ribavirin treatment or supportive care only at Sivas Cumhuriyet University Hospital, Turkey, were analysed. By comparing the frequency of mutations in each group, we found little evidence of an overall mutagenic effect. This suggests that ribavirin, administered at the acute stages of CCHFV infection (at the World Health Organization-recommended dose) is unable to induce lethal mutagenesis that would cause an extinction event in the CCHFV population and reduce viremia.Öğe The investigation of host genetic variants of toll-like receptor 7 and 8 in COVID-19.(2023)Toll-like receptors (TLRs) recognize infectious agents and play an important role in the innate immune system. Studies have suggested that TLR single nucleotide polymorphisms (SNPs) are associated with poor antiviral responses against SARS-CoV-2. Therefore, we aimed to investigate the relationship of TLR7 and TLR8 (SNPs) with COVID-19 disease prognosis. A total of 120 COVID-19 patients, 40 outpatients, 40 clinical ward patients and 40 intensive care unit (ICU) patients were included in the study. TLR7 (rs179009), TLR8-129 C/G (rs3764879) and TLR8 Met1Val (rs3764880) SNPs were genotyped using the PCR-RFLP method. In female patients, individuals carrying AG genotype and G allele for TLR8 Met1Val SNP were found at a higher frequency in patients hospitalized in the ICU than in patients followed in the clinical ward (p < 0.05). In terms of the other two SNPs, no significant difference was found between the groups in females. Furthermore, in male patients, A allele of TLR7 rs179009 SNP was at a higher frequency in patients who have at least one comorbidity than in patients who have no comorbidity (p < 0.05). Our results suggest that TLR8 Met1Val SNP is important in the COVID-19 disease severity in females. Furthermore, TLR7 rs179009 SNP is important in male patients in the presence of comorbid diseases.
