Deri leyişmaniyozu etkenlerinin üretilmesinde farklı bir besiyerinin denenmesi
Abstract
Leishmania türleri ile yapılan çalışmalarda fazla sayıda promastigot elde edebilmek için değişik besiyerleri kullanılmaktadır. Bu amaçla, en uygun besiyerini saptamak için L. tropica promastigotları 5 değişik bifazik, sıvı ve kendi hazırladığımız yeni bir besiyerinde üretilmek suretiyle gelişme fazlan ve parazit yoğunlukları açısından karşılaştınlmıştır. Çalışmamızda en kısa sürede, mümkün olduğunca fazla promastigot üretebilmek amacıyla çeşitli besiyerleri denenmiştir: (i) NNN (tavşan kanlı tuzlu agar), (ii) İnsan kanı içerikli NNN, (iii) RPMI-1640 + %20 FCS + Steril pamuk lifcikleri, (iv) NNN + RPMI-1640 + %20 FCS, (v) "Eğer tavşan kanı bulamazsak veya NNN'den daha kısa sürede ve daha fazla sayıda promastigot üretebilirmiyiz?" sorusunu yanıtlamak için kendimizin oluşturduğu besiyeri. Tüm deneylerde log fazı ortalama l haftada (6-8 gün) oluşmuştur. En iyi üreme, NNN besiyerinin kondansasyon sıvısının üzerine (besiyerinin yatık kısmının uzunluğuna göre) yaklaşık 1-2 mi RPMI-1640+%20FCS besiyeri eklenip, 5-6 saat dinlendirildikten sonra ekim yapılan besiyerlerinde elde edilmiştir (8. günde 135x$10^6$). Bizim hazırladığımız besiyerinde (DPB) de bol miktarda üreme olduğu (8. günde 96x$10^6$) gözlenmiştir. Various media are used to produce promastigote forms of Leishmania species for experimental studies. For this purpose, in order to determine the most appropriate medium, L tropica promastigotes were cultivated in 5 different bi-phase and liquid media and their growth phases and parasite density were compared. All media were inoculated with 1x106 promastigotes. In this study, we have tried to determine the best culture medium giving the shortest period of growth and the highest density of L. tropica promastigotes. These media included NNN (salt agar containing rabbit blood), NNN containing human blood, RPMI-1640+%20 FCS+ sterile cotton fibers, and NNN+RPMI-1640+%20 FCS. We also prepared a medium with which we could obtain growth in the shortest period of time with the highest density of L. tropica promastigotes when we could not find rabbit blood compared to the NNN medium. The parasite density was found to be low in NNN+RPMI-1640+%20 FCS (105x$10^6$ promastigotes/ml). The highest density of L. tropica promastigotes was observed in the NNN+RPMI-1640+%20 FCS medium on the 8th day (135x$10^6$ promastigotes/ml). The third highest density was observed on the 8th day (96x$10^6$ promastigotes/ml) in the culture medium which we prepared.
