dc.description.abstract | Bu çalışmanın amaçları: 1. İki farklı ‘Transforming’ Büyüme Faktörü b-1 (TGF$\beta$-1) dozu içeren ve biyolojik poliyesterlerden yapılmış yeni bir kontrollü salınm sistemi hazırlamak, 2. Bu sistemin periosteal, kortikal ve intramedüller kemik iyileşmesine etkilerini araştırmaktır TGFb-1 kemik osteogenezinde çok iyi bilinen bir sinyal taşıyıcısıdır Bu madde bakteri kökenli, biyo-uyumlu, biyolojik olarak yıkılabilir ve toksik olmayan polihidroksibütirat-co-hidroksivalerat (PHBV) çubuklarına emdirildi. Otuz adet sıçanın her iki tibiasına delikler açıldı ve onbeşer sıçandan oluşan iki grup oluşturuldu. Kontrol amaçlı olarak üçüncü bir grup ise tedavi edilmemiş 5 sıçandan oluşmaktaydı. Aynı boyutlardaki 5.0 ng ve 20.0 ng TGF$\beta$-1 içeren çubuklar sırasıyla Grup 1 ve Grup 2’deki sıçanların sağ tibialarına yerleştirildi. TGFb$\beta$1 içermeyen PHBV çubukları ise fantom olarak sol tibia defektlerine yerleştirildi. Onbeş günluk aralarla sıçanlar sakrifiye edildi ve toplam bir ay sonunda elde edilen spesimenler makroskopik, histopatolojik ve radyolojik olarak değerlendirildi. TGF$\beta$-1 ile muamele edilmiş sıçanlarda, düşük doz içerenlerde daha yavaş olmak üzere, radyografik olarak tam iyileşme gözlendi. Histopatolojik bulgular radyolojik bulguları desteklemekteydi. TGF$\beta$-1 emdirilmiş taşıyıcı materyallerin kemik iyileşmesinde hızlandırıcı etkisi olduğu ve çok az yabancı cisim reaksiyonu oluşturduğu sonucuna varıldı. | en_US |
dc.description.abstract | The aims of this study are; 1. To prepare a novel local delivery system, made of biological polyesters, incorporating two different doses of TGF $\beta$-1, 2. To determine its effectiveness on periosteal, cortical and intramedullar bone healing. Transforming growth factor beta-1 (TGF$\beta$-1), which is a proven prominent messenger in bone osteogenesis, has been adsorbed to microbially synthesized natural, biodegradable, biocompatible, and non-toxic polyhydroxybutyrate-co-hydroxyvalerate (PHBV) rods. Burr-holes were created on both tibias of 30 rats. These were divided into two groups of fifteen subjects. An additional third group, consisting of five rats, was used as non-treated controls. The rods of the same dimensions containing 5.0 ng and 20.0 ng of TGF$\beta$-1 were implanted into the defects of the right legs in Groups 1 and 2 respectively Plain PHBV rods were implanted in the left legs, which was used as phantoms. In fifteen day intervals the rats were sacrificed and at the end of one month, the specimens were analyzed macroscopically, histo-pathologically, and radiologically The radiographs of the subjects treated with TGF$\beta$-1 showed complete healing. However, this was observed to be much slower in low-dose treated implants. Histo-pathologic evalu-ation also supported the radiological findings. It was concluded that the delivery material treated with TGF$\beta$1 have an enhancing effect on bone healing with a minimum foreign body reaction. | en_US |
