dc.description.abstractÖZET Yüksek Lisans Tezi Paraquat' in (Gramoxone) Vicia faba L." da Mitoz Bölünme, Kromozomlar ve DNA Miktarı üzerine Etkileri ŞİFA TÜRKOĞLU Cumhuriyet Üniversitesi Fen Bilimleri Enstitüsü Biyoloji Anabilim Dalı SİVAS Danışman: Yrd. Doç. Dr. Serdar KOCA Bu çalışmada, Paraquat 'm ( Gramoxone) Vicia faba L.( Bakla)' da mitoz bölünme, kromozomlar ve DNA miktarı üzerine etkileri araştırılmıştır. Paraquat' m 10 ppm, 50 ppm ve 100 ppm lik dozları 3, 6, 12 ve 24 saatlik süre ile Vicia faba bitkisinin hem kök hem de tohumlarına uygulanmıştır. Yapılan incelemelerde her iki uygulama sonucunda mitotik indeksin kontrole göre azaldığı saptanmıştır. Ancak Paraquat'in tohuma uygulanması sonucu mitotik indeks daha az etkilenmiştir. Paraquat kromozomlar üzerinde de olumsuz etkiler oluşturmuştur. Paraquat'in köke uygulanması sonucu doz ve sürenin artışına paralel olarak kromozomlarda hasarlar meydana gelmiştir. Paraquat'in 3 ve 6 saat süre ile Vicia faba tohumlarına uygulanması sonucu kromozomlarda hasar oluşmamıştır. Herbisitin 12 ve 24 saat süre ile tohumlara uygulanması sonucu kromozomlarda hasarlar meydana gelmiştir. Tüm uygulamalarda en fazla rastlanan kromozom hasarları anafaz köprüsü ve yapışkanlıktır. Bunların yanı sıra kalgm kromozom, kırılma ve fragment oluşumlarına da rastlanmıştır. Paraquat'in farklı süre ve dozlarla Vicia faba kök ve tohumlarına uygulanması sonucu genel olarak DNA miktarı kontrole göre azalmıştır. Ancak herbisitin 12 saat süre ile Vicia faba tohumlarına uygulanması sonucu DNA miktarı kontrole göre artış göstermiştir. Dr. Serdar KOCA In this study, the effects of Paraquat (Gramoxone) on mitotic division, chromosomes and DNA content were investigated in Vicia faba L. Paraquat dosages of 10 ppm, 50 ppm and 100 ppm were applied to both roots and seeds of Vicia faba plant using periods of 3, 6, 12 and 24 hours. According to our investigation it has been established that mitotic index were decreased in comparison to control in both applications. However, mitotic index was less affected in seeds treated with Paraquat. Paraquat showed some negative effects on chromosomes. Chromosome damages took place as parallel to increases in both dosage and period of Paraquat treatment on roots. As a result of treatment with a period of 3 and 6 hours on seeds of Vicia faba, there were no chromosome damages observed. Chromosome damages were occurred as a result of treatment with a period of 12 and 24 hours on seeds. The most common chromosome damages in all applications of dosage and periods were anaphase bridges and stickiness. Besides, laggard chromosomes, breaking and fragment formation were also found. As a result of the treatment on roots and seeds of Vicia faba with different dosage and period of Paraquat generally decreased DNA content in comparison with control. However, a period of 12 hours treatment with the herbicide DNA content showed an increase in comparison to control.
